Really? I thought Season 1 had the best cliffhanger. The semi hitting the car, leaving all 3 Winchester men bleeding, not knowing if any of them would survive.
As for best, I wish I could have voted for two because I thought the Season 1 and 2 finales (especially if you count AHBL parts 1 and 2 as one) were the best. They are a tie, to my mind.
